mTORi stands for Mammalian target of rapamycin inhibitor, a drug class abbreviation.
Drugs that block mTOR, a regulator of cell growth: sirolimus, everolimus and temsirolimus. They are used to prevent transplant rejection, to treat kidney cancer, breast cancer and tuberous sclerosis complex, and on drug-eluting coronary stents.
Example: Convert CNI to mTORi sirolimus for post-transplant skin cancers.
Usage notes
Sirolimus was discovered in a soil sample from Easter Island (Rapa Nui), which gave it the name rapamycin. Mouth ulcers, high lipids, impaired wound healing and lung inflammation are typical side effects, so it is often avoided immediately after transplant surgery. Unlike CNIs, mTOR inhibitors are not strongly nephrotoxic.
