PD-1 stands for Programmed cell death protein 1, a hematology and oncology abbreviation.
Checkpoint receptor on activated T cells that switches off their activity when it binds PD-L1 or PD-L2, a mechanism tumors exploit to evade immune attack. Antibodies blocking PD-1, such as pembrolizumab and nivolumab, are among the most widely used cancer drugs.
Example: Anti-PD-1 therapy with pembrolizumab every 6 weeks for MSI-H colorectal cancer.
Usage notes
Japanese immunologist Tasuku Honjo discovered PD-1 in 1992, work recognized with the 2018 Nobel Prize. The first PD-1 inhibitors were approved in 2014 for melanoma. Responses are more likely in tumors with high PD-L1 expression, high tumor mutational burden or mismatch repair deficiency.
